Design: Genomic DNA from each individual was digested with Sbf I-HF restriction enzyme was ligated to the unique P1 RAD adapter primers. Adapter ligated DNA fragments from individual samples were pooled and sonicated. DNA fragments between 400-600 bp were extracted using 1% agarose gel. Blunt ends were repaired and A-overhangs were then added using Klenow fragment (3’-5’ exo). P2 RAD adapter was then ligated to the DNA.
